グリコシダーゼの基質認識機構の解析および機能改変

研究概要

Arthrobacter globiformisのglucodextranaseの一次構造を決定し、これがこれまで報告されている他のdextranaseとは相同性が全く認められないが、Clostridium glucoamylaseの一部と高い相同性があることを認めた。このことは酵素のα-1,4結合とα-1,6結合の認識がどの部位でどのようにされているかを究明する足がかりとなるものある。そこで、Clostridium thermoamylolyticumからglucoamylaseの全遺伝子を単離し、大腸菌発現を行うとともに発現酵素の諸性質を解析した。他のglucoamylaseとは異なり、α-1,4結合に良く作用する特徴ある酵素であることを明らかにした。両者の遺伝子を獲得し、それらの発現に成功したことにより、キメラ酵素の作成が可能となった。
澱粉の生合成に関与する枝作り酵素は、アミラーゼの性質を有している。インゲンマメから性質の異なる2種類の遺伝子の単離と各酵素(pvSBE1、pvSBE2)の発現は済んでいた。今年度はこれらのキメラ酵素を作成し、C末端領域がアミロースとアミロペクチンの親和性を決めていることを証明した。また、N末端領域は活性化因子などの調節と関係していることを明らかにした。さらに、このようなキメラ酵素に転移鎖長の相違を見出し、本来一方の酵素が持つ機能の改変に成功した。これらとは別に、N末端領域の延長酵素がデンプン粒結合酵素としての機能を特徴づける事実を証明した
