魚類の急性期応答因子の分子クローニングおよび動態解析

研究概要

細菌感染などの初期に産生量が上昇する急性期応答因子を、魚類の感染防御に応用する方策を検討するために、コイおよびニジマスの急性期応答因子のcDNAクローニングとその発現変動を解析した。真菌由来のβ-1,3-グルカン、海藻由来のアルギン酸ナトリウム、哺乳類の炎症誘発剤であるテルペン油、および魚病菌Vibrio anguillarumを刺激物質として用い、コイの腹腔内浸出細胞およびニジマスの肝臓で発現が上昇した遺伝子をSuppression Subtractive Hybridization法によって解析した。その結果、コイからはpentraxin、serum amyloid Aなど既知の急性期応答因子の他に、ケモカイン、ケモカインレセプター、新規C-タイプレクチン、新規補体B因子アイソタイプ、グリア細胞成熟因子、インターロイキン1β、Pre-B cell enhancing factorなど、約50種の遺伝子が同定された、それらの発現変動がRT-PCRによって確認された。一方、ニジマス肝臓からは、LECT2ケモタキシン、補体B/C2、C-タイプレクチン、フィブリノーゲン、ハプトグロビン、ヘモペキシン、CD59、リゾチウム、ビテロゲニン、プレセレブリン、Toll-like receptor、トランスフェリン、serum amyloid A、C-多糖結合タンパク質、トロンビンなど、36遺伝子が同定された。特に、CD59や補体B/C2など、これまで恒常的に発現していると考えられてきた補体系の成分の発現が、感染刺激にダイナミックに応答していることは、魚類にユニークな特徴であると考えられる。
