孵化直後のニワトリヒナにおける摂食・飲水行動の中枢および末梢制御機構の解明

研究概要	消化管ホルモンの一つであるガストリンと構造が類似しているコレシストキニンの摂食効果を比較したところ、コレシストキニンのペプチド鎖が長い効果は強くなり、ガストリンの効果に近づいた。さらに、アミノ酸のヒスチジンから合成されるヒスタミンにより摂食は抑制されるが、そのヒスタミンは脳内の肥満細胞由来のものであることを認めた。α-黒色素細胞刺激ホルモン(α-MSH)やボンベシンの中枢投与により摂食が抑制されることも示された。上記の事実より、幼雛の摂食抑制因子は哺乳動物のものとほぼ同様であることが判明した。ところが成長が早く摂食量の多いブロイラーと成長が遅く摂食量の少ない産卵鶏の間でGLP-1やα-MSHの阻害剤に対する効果が異なり、遺伝的選抜は脳内の神経ペプチドの発現に差を生じさせていたことが明らかとなった。次いで、摂食亢進因子に関して研究を行ったところ、哺乳類で摂食亢進因子として知られるオレキシン、メラニンコンセントレイティングホルモン(MCH)、ガラニン、モチリンには、幼雛の摂食量を高める効果は認められなかった。モチリン様作用を有するエリスロマイシンの末梢投与によっても摂食量は何等影響を受けなかった。ニューロペプチドY(NPY)とクロニジン(α2-アドレナリン受容体作動薬)には摂食亢進効果が認められ、特にNPYに強い作用があることが判明し、幼雛の摂食亢進因子は哺乳動物に比べ単純化していることが判明した。そのNPYの摂食亢進効果に対し、一酸化窒素合成酵素の阻害剤、α-MSHあるいはボンベシンなどが拮抗的に働くことも明らかにした。浸透圧の調節に関与するアルギニンバゾトシンは、高浸透圧下での放出がオピオイドによって抑えられることや、高浸透圧下では放出のみならず合成も高まることも判明した。
