エンドセリンおよびアンジオテンシン受容体シグナル伝達機構の分子薬理学的解析

研究概要	循環器系の病態生理学的な調節は種々の細胞から遊離されるパラクリンおよびオートクリン調節物質が細胞膜の受容体刺激を介して始動する細胞内情報伝達過程を介して行われるが、ホスフォイノシタイド(PI)代謝共役型受容体とその情報伝達過程は非常に重要な役割を演じている。アンジオテンシンII(Ang II)およびエンドセリン(ET)受容体はPI代謝促進によるIP_3およびdiacylglycerol(DAG)生成を介して細胞機能・細胞肥大やリモデリングなどの調節に関与している。しかし無傷(intact)な心筋細胞でどのように統合的に機能制御を行っているのかについての詳細は現在のところまだ解明されていない。本研究においてはAng IIおよびET-1によるCaシグナル調節を細胞内Caトランジェント(CaT)、膜イオン電流、収縮性の変化を指標に分析した。ウサギ心室筋細胞におけるAng IIによるCaTの増強はNa/HおよびNa/Ca交換機構の選択的阻害薬であるEIPA、KB-R7943および両者の低濃度の組合せで選択的に抑制される。これらの実験結果はAng IIの作用にDAG→PKC→Na/H交換機構活性化→細胞内Na蓄積とアルカリ化→CaT増強・Ca感受性増強→陽性変力作用という細胞内過程が重要な役割を演じていることを示唆する(1)。ET-1の作用もKB-R7943で選択的に遮断され、Na/Ca交換機構の重要性を支持する(2)。またウサギ心筋細胞においてET-1はICaに対して顕著な増大作用を示さず、ET-1の陽性変力作用にICa増強は重要な役割を果たしていないことを示唆し、前述の過程の重要性を支持する(3)。ET-1はむしろβ受容体刺激の存在下で強い抑制作用を惹起する。これらの実験結果はうっ血性心不全などの病態生理学的条件下で、交感神経興奮によりノルエピネフリンが遊離されている状態ではET-1による調節効果は著しく影響を受けることを示唆する。
