三重鎖結合転写因子MAZとp300/CBPによるクロマチンリモデリング

研究概要	三重鎖結合転写因子MAZのDNA結合配列はゲノム上のGCリッチ配列であり、同様の配列にzinc finger転写因子Sp1も結合する。MAZの遺伝子発現を解析する目的でMAZのプロモーター領域を単離同定し、MAZ及びSp1の結合配列を明らかにした。MAZはSp1と同様の配列に濃度依存的に結合する。更にMAZとSp1は共発現によってMAZ自身の遺伝子発現を抑制した。DNA結合実験よりMAZとSp1は共にMAZのGCリッチ配列に結合し、そのDNA結合ドメインは共にzinc fingerであり、互換性があることを見い出した。更にMAZと会合するp300/CBPは発現によりMAZの転写抑制活性を促進することが判明した。この際MAZはヒストンアセチル化酵素であるのに、なぜHDACをリクルートするのかその分子機構を現在解析中である。Sp1はMAZと異なり、メチル化酵素(Dnmt-1)をリクルートし、MAZの転写活性を抑制する。本年度はGCリッチDNAの高次構造とMAZ,SP-1の結合様式の関係を始めて明らかにした。
