Th1/Th2細胞の活性化による微生物に対する感染防御能の増強効果

研究概要	HIV-1、インフルエンザ等で作製した新世代DNAワクチンはCMVプロモーターにウイルスのエピトープをコードさせているDNAを結合させたワクチンである。この場合ウイルス独自のコドンではなく、ヒトで多く使われているコドンを使用しているため、今回、マウスを使用した実験では非常に免疫原性が高いことが判明した。更にユビキチンの遺伝子をCMVプロモーターの後に結合させたところ、免疫誘導能が強まり、インフルエンザのHA、M叉はNPのDNAワクチンでは同じ型のインフルエンザウイルスに強い感染防御能が得られた。緑膿菌については、エラスターゼ、アルカリ性プロテアーゼ、鞭毛抗原及びエクソトキシンAの一部の抗原決定基DNAより成るDNAワクチンは、強い感染防御能があることが判明した。またHIV-1のDNAワクチンは細胞障害性T細胞を誘導する能力が優れているので、現在国立感染症研究所、本多三男博士と30匹のカニクイザルを使用してHIV-1の有効性の検討を行なっている。アデノ随伴ウイルス(rAAV)及び、E1欠損アデノウイルス(Ad)にHIV-1、インフルエンザの一部の遺伝子を挿入し、ウイルスベクターワクチンとして検討を行ない、rAAV及びAdベクターワクチンの強い免疫効果があることが判明した。また、rAAVではインフルエンザウイルスの増殖を阻止する能力が見られた。一方、骨髄より取り出した樹状細胞にE1欠損アデノウイルスワクチンでリポフェクチンと共にin vitroで培養し、感染させた後、この細胞をi.v.によりマウスに注入した。マウスでインフルエンザ及び、緑膿菌に対する感染は阻止することが判明した。以上のことにより、我々の作製している新世代DNAワクチン及び、いくつかのウイルスベクターワクチンは、非常に強い微生物の感染阻止能があることが判明した。
