研究概要

新世代DNAワクチンを使用し、緑膿菌ワクチン、HIV、インフルエンザなどに対するワクチンを作製した。まず、緑膿菌のワクチンについていえば、エラスターゼ、アルカリ性プロテアーゼ、鞭毛抗原及びエクソトキシンA等の複数の抗原決定基をコードするDNAよりなる新世代DNAワクチンは、BALB/cマウスに免疫すると各々の抗原に対する特異的免疫反応が出現し、病原性緑膿菌に対する強い感染予防効果があることが判明した。次にHIV-1では、感染防御に関係する抗原をコードする約18個の遺伝子を直列に結合したDNAワクチンを合成した。18個の遺伝子は各々が約15個のアミノ酸よりなるHIVのenv, gag, polの中和抗体産生部位やCTLエピトープ等をコードしているDNAがヒト型化コドンで、いわゆる多価DNAワクチンである。ウイルスのDNAをヒト型コドン化し、更にヒトの中で発現しやすくしている。強い免疫原性及びHIVとワクチニアウイルス、キメラウイルスの感染実験でもこの新世代DNAワクチンを投与することにより強い感染予防効果があることがわかった。現在この新世代DNAワクチンを使用し、国立感染症研究所、本多三男博士と約30匹のカニクイザルを使用し、有効性の検討を行っている。更にインフルエンザウイルスの場合は、我々は今まで報告が無かったタンパクをコードするMに対するDNAワクチンも作製し、これがインフルエンザの複数の株にも交差防御効果が得られることが判明した。また、インフルエンザについては経皮投与、経鼻投与等でも強い感染防御能が得られることが判明し、更にその実用化に向けて現在検討を行っている。以上我々は今回ヒト型化コドンを使用し、新世代DNAワクチンの作製及びその免疫効果及びその有効性があることをマウスを使用した実験で判明した。
