研究概要

カテキンの抗微生物活性のメカニズムおよびカテキンの抗微生物薬との併用効果について研究を行った。
1.エピガロカテキンガレート(EGCg)は細菌細胞膜や細胞壁傷害作用および細胞壁合成阻害作用を有し、β-ラクタム剤と併用すると、MRSAに対するβ-ラクタム剤の抗菌活性を復活させ、強い相乗効果が発現した。タンパク合成阻害剤、核酸合成阻害剤との併用では相乗効果は見られず、ペプチド系抗生物質では拮抗作用が見られた。この拮抗作用はEGCgとペプチドの結合によると考えられた。また、EGCgはβ-ラクタマーゼ活性を直接阻害することができ、β-ラクタマーゼ産生黄色ブドウ球菌およびMRSAに対して、EGCgとβ-ラクタム剤の併用は相乗効果を発揮した。β-ラクタマーゼが分泌されないグラム陰性桿菌に対して、効果は弱かった。
2.ヘリコバクター・ピロリに対しては、EGCg単独で殺菌作用を示した。クラリスロマイシン高度耐性株に対してEGCgとクラリスロマイシンまたはプロトンポンプ阻害剤を併用すると相加効果が見られた。
3.細胞内寄生菌サルモネラに関しては、EGCgはサルモネラ食食マウスマクロファージの細胞内殺菌能を亢進した。EGCg投与マウスのマクロファージはサルモネラ貧食能と細胞内殺菌能がともに増強した。
4.HIVに関しては、HIVの細胞への吸着後から宿主細胞遺伝子への挿入までの過程においてEGCgの阻害作用が見られ、逆転写酵素やプロテアーゼの阻害が示唆された。また、EGCgは持続感染細胞からのHIV粒子の産生を抑制した。この抑制効果はリボソームに包埋したEGCgやLPSを併用すると増強した。しかし、このEGCgの効果は単球細胞系で特異的におこり、リンパ球では見られなかった。また、EGCgとAZTを併用すると弱いながら相乗効果が見られた。
