B細胞抗原受容体からの情報伝達と機能発現の制御機構

研究概要	本研究ではB細胞の生存と分化におけるB細胞抗原受容体からのシグナル伝達の役割及びその制御機構について、Lynキナーゼを中心とするシグナル伝達及びホスファチジルイノシトル系代謝の制御から検討を加えた。Lynキナーゼ欠損マウスと自己赤血球産生性トランスジェニックマウスの掛け合わせでは、B1細胞の異常増加と分化亢進が見られる。本研究ではB1細胞での抗原受容体からのシグナル伝達の制御、特に負の制御機構に異常を来していると考え、シグナル伝達の負の制御に関わる受容体を検索した。その結果、CD5、CD72、FcγRIIBなどの受容体のチロシンリン酸化にLynが深く関与していること、Lyn欠損マウスB細胞においては、これらの受容体の細胞内ITIMのチロシンリン酸化とITIMへのSHP-1フォスファターゼの会合が見られないことがわかった。さらに抗原受容体刺激後での正常B細胞でのチロシンリン酸化タンパクとLyn欠損マウスB細胞でのチロシンリン酸化タンパクのパターンを比較した結果、Lynキナーゼによって抗原受容体刺激後に強くチロシンリン酸化を受ける新たな分子を見出した。分子量及びその抗原性からLy49分子ファミリーと考えられた。B細胞ではB1細胞上のみにLy49様受容体が発現されており、抗原受容体刺激によりチロシンリン酸化を受け、SHP-1がリクルートしてくることがわかった。このLy49様受容体を抗原受容体からSequesterするとB1細胞での増殖と生存が亢進された。以上の結果は、B1細胞では複数の負の制御を行う受容体が存在しその制御にLynキナーゼが重要な働きをしている事を示している。AktはPI3キナーゼを介して活性化され、細胞の増殖と生存を亢進させる。Lyn欠損B細胞ではAktの異常に高く、しかも長期に続く亢進が見られる。Lynキナーゼは自己抗体産生B細胞の増殖、生存に強い負の制御を行っていることが明らかになった。
