細胞障害性リンパ球による標的細胞破壊機構の解明およびその制御

研究概要

新鮮末梢血T細胞における、様々なマイトージェンやサイトカイン刺激後のTRAIL分子の発現誘導について解析した結果、TRAILの発現誘導にはtype I IFNによる共刺激が必要であることが判明した。Type I IFNは腎癌、黒色腫などの腫瘍に対して、臨床応用されているが、TRAILに対して感受性示した腎癌細胞を標的細胞として末梢血T細胞の標的細胞破壊機構について解析した結果、抗CD3抗体で予め刺激しておくと腎癌細胞を殺傷するようになることが判明した。さらに、IFN-αによる共刺激を加えると抗CD3抗体単独で刺激した細胞と比較して約1.5^〜2倍程度の強い細胞傷害活性が誘導されることが明らかとなった。また、この時、ヒトTRAILに対する中和抗体(RIK-2)を添加するとIFN-αの細胞傷害活性増強効果がほぼ抑制されることが確認された。これらの結果はtype I IFNは抗原を認識してTCR/CD3を介して刺激を受け取ったT細胞に作用し、TRAILの発現を誘導することによりT細胞の腎癌細胞に対する標的細胞障害活性を増強することを示す。(Kayagaki et al.J.E.M.1999)
マウスNK細胞におけるTRAIL分子の発現についても、未刺激時の脾臓由来NK細胞にはTRAILの発現は認められないが、IL-2やIL-15といったサイトカインによってTRAILの発現が誘導されることが判明した。また、IL-2やIL-15によって活性化されたNK細胞がFasLやパーフォリン経路とともにTRAIL経路を介して標的細胞を破壊することも確認された。これらの結果は、CTLのみならずNK細胞もTRAIL経路を介して標的細胞を破壊すること、および、NK細胞においてはTRAILの発現がIL-2やIL-15によって制御されることを示す。(Kayagaki et al.J.I.1999)
