研究概要	本研究ではおもにリンパ球や樹状細胞を標的とする免疫系ケモカインの生理的および病的役割をin vitroおよびin vivoで明らかにすることを目的とする。今年度の成果は以下の通りである。(1)CCケモカインLARCは腸管リンパ組織を被う腸管上皮細胞(follicule associated epithelium)で構成的に発現しており、さらにLPS刺激で一過性に発現が上昇することを見い出した。LARCは腸管免疫に関係する可能性がでてきた。(2)CCケモカインTARCとMDCのレセプターCCR4はヒトの末梢血ではTh2タイプのメモリーT細胞にきわめて選択的に発現していることを明らかにした。TARCやMDCがCCR4を介してアレルギー性炎症に重要な役割をになっている可能性がでてきた。(3)ヒトの染色体7q11.23の領域から新規のCCケモカイン遺伝子を発見し、cDNAクローニングした。このケモカインは新たなCCR3の機能的リガンドであることからeotaxin3と命名した。(4)ヒトの染色体9q13の領域から新規のCCケモカイン遺伝子を発見し、cDNAクローニングした。ILCと名付けたこのケモカインは皮膚に選択的に発現しており、また伝染性軟属腫ウイルスのコードするケモカインとの類似性が高いことが分かった。(5)アトピー性皮膚炎様の皮膚疾患を自然発症するNC/Ngaマウスの病症局所ではTh2型のケモカインTARCとMDCの発現が亢進し、Th2細胞と考えられるCCR4陽性のT細胞が浸潤していることを明らかにした。
