広域ヌクレオチド解析に基づくヒト疾患原因遺伝子の究明と新しいモデルマウスの開発

研究概要	本研究では,我々が決定した、ヒト第6染色体短腕6p21.3に位置するHLA領域の極めて精度の高いゲノムシークエンシングの成果(1.8Mb)の成果をもとに、2つのヒト疾患(ベーチェット病および尋常性乾癬)について,その遺伝的素因となる原因遺伝子の候補を同定し、それぞれの疾患モデルとなるマウスを作製することを目的とした。前年度までに、患者および健常人のマイクロサテライトマーカーと単一ヌクレオチド多型に関する解析から、ベーチェット病に関してはほぼHLA-B51アレルが、尋常性乾癬ではSEEK1という遺伝子が疾患の発症に深く関わっていることを示すことができた。本年度は、ベーチェット病疾患モデルマウスの開発と尋常性乾癬原因候補遺伝子の解析を中心に研究を進め、以下の成果を得た。 1.ベーチェット病疾患モデルマウスの開発では、HLA-B51遺伝子とヒトβ2マイクログロブリン遺伝子をもつ2重トランスジェニック(Tg)マウスを交配により作成していたが、このマウスでは、HLA-B51の細胞表面上への発現がもとのHLA-B51 Tgマウスに比べ、飛躍的に増大していることを確認した。さらにマウスβ2マイクログロブリン遺伝子欠損マウスと交配することによりHLA-B51遺伝子の発現に関しては完全にヒト型のマウスを作製する事に成功した。 2.尋常性乾癬原因候補遺伝子の解析。 (1)昨年度までに同定したSEEK1遺伝子について、さらに患者および健常人の塩基配列を解析し、単一ヌクレオチド多型を決定した。 (2)SEEK1遺伝子を中心に患者の患部、非患部あるいは健常者皮膚での遺伝子発現をマイクロアレイ法で比較し、患者の患部で発現が大きく変化する遺伝子を特定した。 (3)SEEK1遺伝子のTgマウスを作製し、数匹のFounderマウスを得る事ができた。
