研究概要

リンパ造血器細胞の文化増殖における転座切断点領域遺伝子卯の役割を検討した。
MLL遺伝子は多様な転座相手遺伝子を有するが、t(9;11)転座は骨髄球性白血病に強く相関して認められ、t(4;11)転座はリンパ性白血病に強い相関が認められるため、MLL遺伝子は細胞分化に関して、重要な役割を担っていることが推測される。そこで、白血病細胞におけるMLL遺伝子転座を代表する変異MLLであるMLL-Zf(-)を安定発現する導入細胞と、ベクターのみの導入細胞で、発現に差のある遺伝子をRDA(representational difference analysis)法で解析した。MLL-Zf(-)導入細胞でG-CSF存在下で発現が増強する遺伝子として、あるsubtypeのPI-3 kinase遺伝子を単離した。別の変異型MLLであるMLL-LTG9の導入細胞でも同じ遺伝子の発現増強が認められ、標的遺伝子であることが輝かとなった。
粘膜関連リンパ組織(MALT)リンパ腫に関与するt(11;18)転座の本態は細胞死を抑制するAPI2遺伝子と18番染色体上の新規遺伝子MALT1が融合遺伝子を作ることであることを明らかにしたが、MALT1遺伝子の役割を蛋白レベルで解析するために、MALT1遺伝子を大腸菌に発現させ、リコンビナント蛋白を精製し、4種類のモノクローナル抗体を作成した。現在、組織レベルでの検討をおこないつつある。
我々は、び慢性大細胞型B細胞性リンパ腫(DLBL)に関連するBCL6転座を解析したが、複数の疾患単位からなるDLBLのなかに、特徴的な病型を見出すことはできなかった。しかし、CD5+DLBLが四語の悪い疾患単位を形成することを見出し、多数症例による解析にて、より明確となった。この分化段階を代表するマーカーを見出すため、cDNA array法にて検討しつつある。
