研究概要	脂肪細胞の多寡は肥満や糖尿病の発症に深く関与する。細胞内にトリグリセリド(TG)を蓄積しているが、その分解の調節に与える酵素としてホルモン感受性リパーゼ(hormone-sensitive lipase;HSL)が知られる。HSL の脂肪分解における役割を調べるために、HSL を欠損するマウスをジーンターゲティングによって作成した。HSL ノックアウトマウス(HSL-/-)は、一見正常であり、体重増加曲線を調べる限り、肥満はきたさず、体の各部位の脂肪組織重量の増加はみとめられなかった。ところが、傍精巣上体脂肪組織や褐色脂肪組織を顕微鏡的に調べると、脂肪細胞の肥大が観察された。DNA あたりの TG 含量の増加も認められた。β-アドレナリン受容体刺激活性を有するイソプロテレノールを静脈注射すると、血液中の遊離脂肪酸(FFA)とグリセロール濃度が増加した。白色脂肪組織から脂肪細胞を分離培養した系でイソプロテレノールを添加しても、野生型に比較して弱いながら、HSL-/-においても培養液中の FFA とグリセロール濃度の上昇が観察された。このことは、HSL 以外にも細胞内のTGを加水分解する酵素が存在し、しかもこの酵素はホルモン感受性であることを示している。 TG リパーゼ活性を測定したところ、HSL-/-の褐色脂肪組織においては野生型と同レベルのTG リパーゼ活性が認められた。一方、HSLのもうひとつの基質であるコレステロールエステルの加水分解活性は、HSL-/-においてほぼ完全に欠損していた。HSL-/-の白色脂肪組織は野生型に比較して低値ながら、十分量の TG リパーゼ活性が認められた。これらの事実から、脂肪細胞には HSL とは異なる新たな TG リパーゼが存在することが、始めて明らかにされた。
