EX VIVO遺伝子導入を行った異種心筋細胞移植に関する研究

研究概要	1.胎児心筋細胞移植によって心機能の向上がすでに報告されているものの、心筋梗塞領域へドナー細胞を生着させるには、虚血が原因で困難であるといわれている。そこでその問題を解決すべく、強力な血管新生因子であるVEGF165導入することを計画し、それを発現するplasmidベクターを作成した。そしてfibroblastにpolyamidoamine dendrimerを利用してin vitroにて遺伝子導入し、その発現を確認するとともに、EBV-Episomal vectorではより強く発現することを確認した。しかしprimary cultureしたfetalのcardiomyocyteでは、polyamidoamine dendrimerによる細胞毒性が強く、新たな遺伝子導入法としてBiomaterialであるatelocllagenを用いた遺伝子導入法を試みている。(谷口) 2.正常ラットを開胸し、生体位心臓の左心室心尖部よりコンダクタンスカテーテルとミニセンサーを挿入し、心臓の拍動に伴って起こる圧容積ループを連続記録した。大動脈の狭窄を行い、心臓に後負荷をかけ、収縮末期容積関係を求めた。ウレタンとネンブタール麻酔下で、それぞれのパラメータの正常値を求めた。心筋梗塞モデルについては現在検討中である。(高木) 3.安全性の高い非ウイルス性ベクターによる、心不全の遺伝子治療を目的とした。我々はまず、EBV-プラスミドを超音波ガイド下に心筋内注入し、高発現が得られることを見いだした。次にβ2-AR遺伝子を心筋症ハムスターに導入し、経時的に超音波にて心機能を評価した。その結果、β2-AR遺伝子を導入したハムスターは、約40%の心拍出量の増加を示し、イソプロテレノール負荷によりさらに約1.5倍に増大した。一方正常ハムスターでは、このような変化は認められなかった。したがって、非ウイルス性ベクターによる、心不全の遺伝子治療の可能性が示唆された。(松田)
