痛み刺激の予測により誘導される不快がサルの大脳皮質活動に与える影響

研究概要	本年度は前年度の研究で明らかにされた痛覚受容性神経細胞が分布する領域に2%lidocaneを1-4μl微量注入し、痛覚受容性神経細胞活動を不活性化し、光刺激強度変化弁別速度に対する影響、警告信号に対する影響について行動学的に検索する予定であった。しかし、現在サルのトレーニングにトラブルが生じ、未だに温度刺激強度変化弁別課題を訓練中であり、麻酔実験には至っていない。この様な課題を訓練する場合、サルによって非常に早く課題をマスターできるものとそうでないものが出てくる。これまでに、2頭について従来と同様の課題について訓練を行ってきたが、これらのサルについては全く訓練の成果が現れていない。そのため、新たに2頭を追加して訓練を行っている。その内の1頭については、ほぼ訓練が終了した。また、このサルについては前帯状回の麻酔の影響をより明確にするため、可逆的な痛覚過敏を起こさせることも計画した。現在までに、この可逆的痛覚過敏モデル動物に関するプレリミナリーな実験結果が得られているので報告する。この痛覚過敏モデルは顔面皮膚にカプサイシンを塗布することにより作成する。カプサイシン処理動物においては侵害的(46℃)および非侵害的(44℃)熱刺激共に、有意に逃避行動の増加を誘発した。また、刺激強度弁別速度はカプサイシン処置により有意に短くなった。この様な可逆的痛覚過敏モデルを用いることにより、前帯状回の機能不全による、サルの温熱刺激弁別速度への影響および温熱刺激に対する逃避行動の出現率に対する影響を、より明確に検出することが出きると考えられる。来年度はさらにこのサルを用いて、前帯状回の麻酔および微小刺激の影響について詳細に検索していく予定である。
