研究概要

本研究の目的は、キャピラリーやマイクロチップ中で行う無担体電気泳動の特性を明らかにし、種々の物質の分析へ応用することにある。過去2年間の研究により、これらの目的のうちキャピラリー/マイクロチップ電気泳動の特性解明の部分については、期待以上の成果を上げることができたが、それらの成果の中で分子間相互作用解析への応用については特に目立つ進展があったので、本年度はこの応用研究の超微量化を進め、生体内分子間認識の研究へと発展させることができた。一方、装置や技術面においてキャピラリー電気泳動に関する部分はすでに完成の域にあるが、マイクロチップ電気泳動はゲノム解析の領域を除き未だ発展途上にある。そこで本年度はマイクロチップ電気泳動の高度な装置化と、新しい領域、特に糖質分析への展開を中心に研究を続行した。これらの成果をまとめると、下記のとおりである。
1 レーザー励起蛍光検出キャピラリー電気泳動による分子間相互作用解析の超微量化:nMレベルの低濃度で糖タンパク質糖鎖と機能性タンパク質の間の相互作用を解析することができた。
2 シングルポイントレーザー励起検出方式マイクロチップ電気泳動による糖分析法の開発:迅速簡便な方法により糖質試料を蛍光標識してマイクロチップ電気泳動分離し、レーザー励起蛍光検出することにより、femtomolで秒単位の超微量高速糖質分析を達成することができた。
3 全流露同時紫外部吸収検出方式によるマイクロチップ電気泳動の可能性についての検討:レーザー励起蛍光検出においては検出波長が特定されるため適用範囲が限定されるので、これを克服できる汎用検出法として全チャンネル紫外部吸収を同時検出する方式についても検討し、1-フェニル-3-メチル-5-ピラゾロンにより誘導体化すれば高速で糖質の簡易分析を行うことができることを明らかにした。
