肝障害抑制因子としての一酸化窒素の役割に関する研究

研究概要	肝臓の障害時に誘導される一酸化窒素合成酵素(iNOS)の役割は従来からその二面性が報告されている。障害を引き起こす要因(酸化ストレス、アルコール、熱、肝毒物)によって、産生される一酸化窒素(NO)は細胞保護あるいは逆に細胞毒性に働く。しかし、その産生制御や作用のメカニズムに関しては不明な点が多い。炎症性サイトカインのインターロイキン1β(IL-1β)により発現誘導するiNOSに関して以下のような知見を得た。 1.低酸素は肝細胞のIL-1β/iNOSの誘導をその転写レベルで抑制する(Hepatology2000;32:1037-1044)。一般にiNOS誘導は低酸素により活性化されることが報告(マウスマクロファージ)されている。低酸素は肝細胞では逆にiNOSプロモーター/ルシフェラーゼ(1.4 kbase)のtransactivation活性を抑制しNO産生を阻害した。この阻害は転写因子NF-κBの活性化(核移行およびDNA結合)やプロモーター領域に存在するhypoxia-response elementには無関係であった。低酸素を介したiNOS誘導抑制の新しいメカニズムの存在が考えられる。 2.NF-κBのvicinal dithiol残基はプロモーターのκB部位への結合に必須である(J Biol Chem 2000;275:4369-4373)。IL-1βのiNOS誘導にはNF-κBの活性化が必須であるが、十分ではない事が良く知られている。Vicinal dithiolに特異的に結合する酸化フェニルアルシンを用いて、NF-κBはiNOSプロモーターのκB部位への結合に隣接するチオール残基の存在が重要な役割をはたしていることを明らかにした。このチオール残基の作用を制御する因子が上述の十分条件に関与する可能性がある。
