研究概要	1)新規の候補遺伝子として、BLyS, BCMA, NKG2C, CD94の多型スクリーニングを施行した。BLYS遺伝子には、プロモータ領域の4個所のSNP、翻訳領域に1個所のアミノ酸置換を伴う稀な変異を見出した。プロモータ領域のSNPは、3種のハプロタイプを形成して存在すること、うち一つが、単離した末梢血単球中のBLyS mRNA量と関連することを見出した。これらのプロモータ多型と、RAやSLE感受性との有意な関連は検出されなかったが、SLEにおいて、抗Sm抗体陽性者にプロモータ活性の高いアリルが多い傾向が観察された。また、アミノ酸置換を伴う変異は、ごく少数であるが、RA患者のみに出現することが観察された。同様に、BCMAにも4種のハプロタイプを見出したが、疾患との関連は検出されなかった。 2)NKG2C遺伝子の変異解析の過程で、PCRによるNKG2Cの増幅が不可能な個体が存在することを見出し、Southern blotting, RT-PCRの結果、NKG2C欠失アリルの存在が示された。NKG2Cホモ欠損者は、一般集団中に4.3%存在することから、欠失アリルの頻度は21%,一般集団中のヘテロ接合体の頻度は33%に達すると推測された。この欠失アリルと、RAやSLEとの関連は検出されなかった。また、CD94については、一個所のSNPを検出したが、RA, SLEとの有意な関連は認められなかった。 3)前年度にSLEとの関連を見出したFCGR2B-I232Tについて、機能解析を施行するため、ヒトFcγRIIb欠損B細胞株に遺伝子導入を行い、通常型・変異型それぞれを発現するB細胞株を作製した。 4)前年度までにRA滑膜における強発現を見出したIDファミリー遺伝子群について、滑膜における機能的役割を探るため、培養滑膜細胞、血管内皮細胞に遺伝子導入を行った。
