研究概要

系統の異なる6種のマウスで、合成女性ホルモン、ジエチルベスタノール(DES)を用いて検討した。
1.近交系マウス6種に、交配後10日目より7日間DESを腹腔内投与し、出産10-11日後の子マウスの脾、胸腺、副腎、腎、胸骨、肺組織の一部を検体とした。心肺は、Carnoy's液で固定した。
2.全系統で、DES 0.5-1.0mg/kgで流産や出血を伴う異常分娩が、0.1-0.5mg/kgでは胎児は正常に分娩されるがそのほとんどが死産であり、0.01-0.02mg/kgで正常分娩とその後の良好な成育が確認された。
3.DES(0.01mg/kg)投与マウスから出生した生後10日目の子マウスの所見を下記に示す。
(1)全系統の対照群とDES群で、心臓及び肺の形態学的異常、奇形等は認められなかった。(2)エンドセリン(ET-1)とその受容体(ETA)のmRNA発現量をC3HとBALBの対照群とDES群で比較した。C3Hでは腎、副腎、胸骨いずれでもET-1,ETA mRNA発現が認められ、かつその発現量に差を見とめなかった。それに対して、BALBでは両群の腎でET-1,ETA,mRNA発現量に差を認めないが、副腎と胸骨ではDES群でETA mRNA発現の減少が確認された。ET-1 mRNAもDES群の両組織で僅かながら減少していた。(3)全系統で、対照群に比しDES群で体重が大であった。胸腺重量及び総細胞数に対するDESの影響には、系統差が認められた。全系統で、脾臓重量と総細胞数がDES群で明かに大であった。細胞性免疫に関しては、全系統のDES群で脾リンパ球のB細胞の割合がやや高い傾向を示したが、T細胞サブセット(CD4/CD8)の割合には明確な差を認めなかった。胸腺細胞の90%以上がTh1.2陽性であり、両群間に差を認めなかった。
以上の結果から、心肺組織のマクロ的形態観察では胎児期に受けたDESの影響は明確ではないが、系統によってDESが細胞性免疫、循環器系、骨代謝系に影響を与えている可能性が示唆された。現在例数をさらに蓄積し、系統差を明確にすると共に、肺構築への影響を細胞および遺伝子レベル、機能レベルで検討中である。
