糖鎖自動合成システムを用いる複合糖質の効率的合成研究

研究概要	糖転移酵素群による効果的な糖鎖伸長反応とその迅速な評価法を確立する。自動合成装置開発との関連において鍵となる「新しい糖転移酵素」の発見・単離、精製・活性(基質特異性)測定の手法についてはこれまで、主として放射性同位元素でラベル化された糖ヌクレオチドを糖供与体とする手法が一般的となっているが、操作の汎雑さや危険性から、より簡単で安全な新しい方法論の確立が急務となっている。糖鎖に二種類の異なる蛍光性基を位置特異的に導入して二重プローブ型の基質やリガンドを合成できれば、この二種類の蛍光プローブ間で観察できる「蛍光エネルギー移動現象」により低濃度(nM)における超高感度での相互作用測定が可能となる。この原理を利用して申請者は全く新しいタイプの「糖脂質分解酵素セラミドグリカナーゼ」に対する高性能蛍光基質の合成に成功し、これまでの皆無であった糖脂質分解酵素の簡易アッセイ法を開発した実績がある。すなわち、糖脂質分解酵素のみならず、糖ペプチド分解酵素であるエンドグリコペプチターゼ(Anal. Biochem.,1995年)やα-アミラーゼ(Biochem.Biophys. Res. Commun.,1996年)のようなendo型の分解酵素についてあまねく広く応用可能な方法論であることを実証してきた。(Tetrahedron Asymmetry.,1994年等) そこで、初年度はまず、この原理を「糖転移酵素」による生合成反応の迅速・定量的解析システムの開発に応用することを計画した。まず糖供与体の特定部位に蛍光性プローブFAを化学的に導入した。次いで、このプローブとの間での良好なエネルギー移動の期待できる蛍光プローブFBを糖受容体の還元末端側に付与させた化合物を合成した。この際、プローブにはナフチル基とダンシル基のようなForster distanceを成立させる組み合わせを選択した。シアリルトランスフェレース等の糖転移酵素による糖鎖のカップリングが起こった際にのみ蛍光エネルギー移動が観測されるのでこれを指標として速度論パラメーターKm, V max等を簡単に決定することが可能となった。さらにこの手法があらゆる糖転移酵素によるグリコシド形成に応用できることを実証するためフコシルトランスフェラーゼやガラクトシルトランスフェラーゼなどの主な糖転移酵素の反応系で用いられる糖供与体・受容体プローブを設計・合成しこれらを評価した。本法が確立できたので糖鎖生物学および関連する領域で広く用いられる事が予想される。
