エンドサイトーシスとエキソサイトーシスにおける小胞輸送の新しい制御因子の機能解析

研究概要	真核細胞のエンドサイトーシスとエキソサイトーシスにおける小胞輸送を制御する分子と考えられているHrsおよびその結合蛋白質(Hbp)について解析を行い以下の結果を得た。 1.EGF受容体およびPDGF受容体とGFPとの融合蛋白質を作成し、これらの受容体の細胞内動態におけるHrsの関与について解析した。融合蛋白質をPAE細胞に導入し、安定に発現する細胞株を取得した。これらの細胞にHrsおよびHrsの種々の変異体を導入し、受容体とGFP融合蛋白質の細胞内動態をGFPの蛍光発色により観察した。その結果、増殖因子で刺激しない細胞において野生型のHrsを導入した場合、受容体が初期エンドソームに蓄積した。しかしZnの結合能を失ったFYVEフィンガードメインをもつHrsを導入した場合では蓄積は観察されなかった。これらの結果はHrsは内在化した増殖因子受容体の初期エンドソームからの移行の選別に役割を果たしていることを示唆している。 2.HrsとHbpが、肥満細胞における分泌小胞からの物質分泌において機能しているか否かについて解析を行った。Hrsとの結合ドメインであるcoiled coil motifあるいはSH3ドメインを欠損するHbpを、肥満細胞株RBL-2H3に安定に発現し、脱顆粒に対する影響をβ-ヘキソサミニダーゼの遊離を指標にして調べた。その結果、IgE受容体による脱顆粒が欠損変異体発現細胞において顕著に阻害された。しかしカルシウムイオノフォア処理による脱顆粒はあまり阻害が見られなかった。したがってHbpの欠損変異体はIgE受容体による脱顆粒反応に対してドミナントネガティブ分子として作用することが明らかになった。またHbpはHrsおよびSH3ドメイン結合蛋白質と相互作用することによりこの脱顆粒反応において機能することが示唆された。
