基底膜型細胞接着分子ラミニン5の構造と機能

研究概要	基底膜型細胞接着分子ラミニン5(以下LN5)はα3、β3、γ2鎖からなるラミニン分子で、インテグリンα3β1、α6β4などの受容体に結合することにより非常に強い細胞接着活性と運動促進活性を示す。本研究ではLN5の構造と機能の関係を明らかにするため、α3鎖のカルボキシル末端領域に存在する5個の球状ドメイン(G1〜G5ドメイン)の機能を調べた。ラミニンα3鎖を発現さず、β3鎖とγ2鎖を発現しているヒト線維肉腫細胞株HT1080に、G5からG1まで順番に欠失させたα3鎖の変異体のcDNAを導入することにより、Gドメインを部分的に欠失したLN5を強制的に産生させた。その結果、G5およびG4-5を欠失するLN5は強い接着活性と運動活性を示すが、G3ドメインを欠失させると両活性が殆ど消失することが明らかになった。また、全てのGドメインを欠失させると殆どLN5が産生されなかった。一方、多くの細胞でα3鎖のG3とG4の間で内在性のプロテアーゼによる切断が起こって、G4-5を欠失するLN5が分泌されること、およびG4-5断片はLN5と遊離して分泌されることが明らかになった。これらの結果から、インテグリン結合配列を有するG2ドメインはそれ単独でインテグリンとの高い親和性を示すことはできず、G1、G3および他のドメインとの間で形成させる立体構造がインテグリンとの結合に重要であることが示唆された。プロテアーゼにより切断されたG4-5断片はLN5と協調的に、あるいは独立して細胞に作用し、何らかの影響を与えている可能性が考えられる。一方、上記のHT1080細胞はLN5とともにラミニン6(α3、β1、γ1鎖)を分泌することが明らかになった。LN5におけるβ3鎖とγ2鎖の役割を明らかにするために、現在両ラミニン分子の性質の比較を行っている。
