モノオキシゲナーゼの構築-P450酸化活性種のタンパクモデルによる検証

研究概要

本年度の研究では、ミオグロビンミュータントに芳香族基質であるトリプトファンを活性中心近傍に導入することを目的に、43番目のアミノ酸をトリプトファン、64番目のアミノ酸をロイシンに置換したミュータント(F43W/H64Mb)を合成した。比較として64番目のアミノ酸をロイシンに置換したミュータント(H64Mb)も併せて合成した。H64Lミオグロビンは過酸との反応で、P450の酸化活性種と考えられている高原子価活性種であるcompound Iを数百ミリ秒の時間スケールで生成する。これに対して、F43W/H64Lミオグロビンミュータントは同様の条件で反応が完結するものの、生成物はcompound Iではなく、トリプトファンの芳香環が水酸化された。この水酸化反応はESI-Massの時間分解測定で追跡が可能であり、反応の詳細を検討した結果、トリプトファンが最終的に6電子酸化を受けていることが明らかになった。
酸化を受けたミュータントは加水分解酵素によって分解し、反応部位を含むオリゴペプチドのNMR測定と、MALDI-TOF Mass測定によるシークエンスの確定によって構造を解明した。
本研究で見出された反応は、これまでタンパクモデルでは達成できなかった芳香環の水酸化反応であり、比較として用いたH64Lミュータントの反応性などの比較から、compound Iによる水酸化反応であると結論された。
