研究概要	1.腸管に特徴的な免疫応答のin vitro実験系の確立これまでに、パイエル板由来樹状細胞がIgA産生を増強することが知られるインターロイキン(IL)-6を高産生することを見い出した。そこでこの細胞を用いたIgA産生誘導のin vitro実験系を構築することを試みた。その結果、パイエル板樹状細胞を抗原+T 細胞+B細胞の培養系に添加することにより、IgA抗体の産生が誘導された。このことから、パイエル板樹状細胞が腸管におけるIgA抗体産生に関与することが示唆され、この系を用いてこのIgA抗体産生誘導機能に対する食品成分の作用を解析することが可能と考えられた。 2.腸管免疫細胞を利用した評価系による食品成分の腸管免疫応答への作用の解析 (1)これまでに小腸上皮細胞のIL-7の産生、およびTGF-βの産生がヌクレオチドの経口投与により増強されることを見い出した。そこでヌクレオチドによる小腸上皮細胞のサイトカイン産生制御機構について解析した。BALB/cマウス由来の小腸上皮細胞の初代培養系にヌクレオチドを添加したところ、IL-7の産生が増強された。このことより、ヌクレオチドは直接小腸上皮細胞に作用してIL-7産生を増強させていることが明らかとなった。一方小腸上皮細胞の初代培養系にヌクレオチドを添加してもTGF-β産生は増強されなかった。このことより、IL-7とTGF-βでは、ヌクレオチドにより産生増強に至るメカニズムが異なることが示唆された。(2)ラップフィルムの可塑剤アセチルクエン酸トリブチル(ATBC)の投与により経口免疫寛容誘導が部分的に解除されることが示された。
