研究概要

本研究は、アニオン性薬物の体内動態において中心的な役割を果たす有機アニオントランスポーターOATファミリーの未同定のメンバーをさらにクローニングし、それも加えたOATファミリーのヒト型トランスポーターの機能解析、及び臓器での発現の局在の検討を行ない、OATファミリーの薬物動態における意義を評価することを目的とする。平成13年度は、既に得られているOAT1〜4の配列を基に、ヒトゲノム塩基配列データベース及びEST(expressed sequence tag)データベースを検索し、2種のOAT類似新規部分配列を同定した。各々の塩基配列に相当するプローブを用いて全長cDNAを単離し、ツメガエル卵母細胞に発現させ機能解析を行い、OATファミリーの新規メンバーURAT1及びOAT7を明らかにした。URAT1は、腎臓に特異的に発現し、その特異抗体を用いた免疫組織化学的検討により、近位尿細管管腔側膜に存在することが明らかになった。URAT1は、尿酸、乳酸、ピラジンカルボン酸、ニコチン酸等を選択的に輸送する有機アニオン交換輸送体であった。ヒト URAT1遺伝子は、10個のエキソンからなり、検討した低尿酸血症患者において、トリプトプァン258のストップコドンへのホモの変異が確認された。これにより、URT1はヒト腎尿細管からの尿酸の再吸収おいて中心的な役割を果たすトランスポーターであることが明らかになった。OAT7は、肝臓特異的に発現し、免疫組織化学的検討では類洞側膜に存在していた。OAT7は、エストロン硫酸、デヒドロエピアンドロステロン硫酸をはじめとする硫酸抱合体を輸送し、腎、胎盤型のOAT4と類似の基質選択性を示した。OAT7は、肝細胞内で生成した薬物、外来性異物あるいはステロイドホルモン等の硫酸抱合体の血液側への出口として機能していると考えられる。
