研究概要	本年度の研究においては、プロトオンコジーン産物、H-Ras及びc-Mycによるリンパ球の細胞接着機構の解析を行った。活性型H-Ras(H-Ras^<V12>)をマウスプロB細胞株BAF-B03に遺伝子導入により発現させると、VLA-4(very late antigen-4)(及びVLA-5)、VLA-6の活性化が誘導され、その結果フィブロネクチン及びラミニンへの接着が亢進すること、またH-Ras^<V12>とc-Mycが協調的に作用することにより、VLA-4インテグリンの活性化に加え、VLA-4のリガンドであるVCAM-1(vascular cell adhesion molecule-1)の発現が誘導される結果、ホモフィリックな細胞接着が引き起こされることが明らかとなった。これまでに、リンパ球系腫瘍細胞のin vivoでの増悪化(腫瘍の浸潤・転移)に伴い、in vitroにおいてホモフィリックな細胞接着の増強が認められることが知られているので、今回我々が見出した知見は、H-Rasとc-MycによりBAF-B03細胞がin vivoにおける浸潤・転移能を獲得した可能性が考えられる。実際、我々はH-Ras^<V12>とc-Mycを構成的に発現するBAF-B03細胞が、ヌードマウスにおいて腫瘍形成に加え、浸潤・転移を引き起こすことを見出している。更に、H-Ras^<V12>とc-Mycの協調作用により、癌抑制遺伝子産物であることが報告されているα6インテグリンの発現が抑制されることが明らかとなった。この知見は、癌(原)遺伝子産物の協調作用により癌抑制遺伝子産物の発現が抑制されるという新しい機序を示していると考えられる。
