血管より産生される新たな分泌タンパクの生理活性解明とその臨床応用法の開発

研究概要	【目的】A55改めDANCEと名付けられた新規の分泌蛋白遺伝子の発現をマウス胎児を用いたin situハイブリダイザーションにより検討したところ、形成過程にある動脈、神経提細胞由来組織である心臓内隆起・心流出路などに強い発現を認めた。特に後者では、上皮-間葉転換機序に関わっていることが示唆され、組織の構築が求められる再生医学への近年の社会の要望の高まりの中で注目される。成体マウスの大動脈では動脈硬化の好発部位である動脈分岐部に強い発現が認められ、高脂質食下のLDL受容体欠損マウスを用いた検討で、動脈硬化病変に一致して発現が上昇していることが確認された。また、ラットの頸動脈バルーン傷害モデルを用いた発現の検討からは、新生内膜病変の血管平滑筋・内皮細胞が病的増殖を停止し始めた部位・時期にその発現が最強となっている。以上の知見からDANCEは病的な細胞増殖を抑制する可能性が示唆された。これは、我々がDANCEの類縁遺伝子群として提唱したDASファミリーの一員であるS1-5蛋白も線維芽細胞の増殖停止に伴い発現が上昇することと併せ、興味深い事実である。臍帯静脈内皮細胞を用いた検討からは、DANCEがインテグリンを介して内皮細胞と結合することが示唆された。また、遺伝子欠損マウスの作成にも成功し、心血管系を含む複数の奇形を認めており、現在その解析を進めているところである。このように、DANCEは発生・病態形成といった幅広い場面で血管細胞を中心とした各種の細胞に作用を持つことが明らかとなった。特に臨床応用の観点からは生理的・病的血管のリモデリングにおける細胞増殖抑制に関わっている点が、分泌蛋白としてのリコンビナントDANCEあるいはその構造類似合成物の投与による経皮的冠動脈形成術術後の再狭窄予防・動脈硬化の予防などへの道を切り開く可能性を有するものとして期待される。
