顎顔面領域における高効率かつ部位特異的な遺伝子導入法の開発

研究概要

平成12年度は、[1]遺伝子発現制御レトロウイルスベクターの改良と[2]高電圧パルス穿孔法による遺伝子導入条件の検討ならび[3]微小電極を用いた微小領域への遺伝子導入実験を行なった。
[1]前年に引き続き、ニワトリレトロウイルスベクターRCASにCre-loxPシステムの応用により導入遺伝子の発現制御を試みた。本年は、感染力の弱いsubgroup Eのニワトリレトロウイルスベクターにlox-P配列を組み込み、感染力の強いsubgroup BのウイルスベクターにCre recombinaseを組み込んだ。レポーターにはGFP遺伝子を用いた。ウイルスフリーのSPF卵より線維芽細胞を分離して、GFP遺伝子を組み込んだウイルスを感染させた。その後、さらにrecombinaseを組み込んだウイルスを感染させた。感染一週間後に、蛍光顕微鏡にて細胞内でのGFP発現を観察した。GFP遺伝子のみを導入した細胞では、70%以上がGFP発現が検出された。一方、GFPおよびCre recombinase遺伝子を導入した細胞では、GFPの発現はほぼ完全に消失した。[2,3]また、高電圧パルス穿孔法による遺伝子導入条件の検討を行なった結果、遺伝子を導入する組織の抵抗値によって条件は、若干異なるものの、20-60V/cmの電圧で50msec,2-4回のパルスの範囲で至適であった。さらに、同方法の実用化を検討する目的で、マウス培養歯胚の微小領域にソニックヘッジホッグ(Shh)遺伝子を導入する実験を行なった。Shh遺伝子がエナメルノットに導入された歯胚では、遺伝子導入部位での同遺伝子発現が確認された。また、同遺伝子に対するアンチセンスオリゴによる歯胚発生阻害作用を一部レスキューした。
