癌の骨転移関連遺伝子のクローニングとその機能解析

研究概要

1.乳癌の骨転移におけるTGF-βおよびIGF-1シグナルの役割
骨組織中には、TGF-βおよびIGF-1が豊富に存在する。従って、乳癌が骨に転移し骨吸収を促進する際に、骨組織からTGF-βおよびIGF-1が溶出され、乳癌の増殖および骨吸収活性に影響を及ぼすと推定される。そこでIGF-1を乳癌細胞MDA-231に添加したところ、その増殖が著明に促進された。IGF-1レセプターのdominant-negative変異体をMDA-231に過剰発現したところ、その骨転移形成および骨吸収活性は著明に抑制された。一方、TGF-βは、MDA231細胞の増殖には効果を示さなかったが、骨吸収促進因子であるPTHrPの産生を著しく促進した。さらにMDA-231にTGF-βを作用させると、TGF-βの細胞内シグナル伝達因子であるSmad2,Smad3およびSmad4を活性化し、その標的遺伝子の転写活性を促進することが明らかとなった。またSmad4を欠損する乳癌細胞は、骨転移を来たさないことが明らかとなった。以上の結果より、乳癌細胞の骨吸収過程におけるTGF-βおよぴlGF-1の重要性が示された。
2.乳癌の軟組織転移に対するビスフォスフォネートの作用
癌原発巣から転移の成立過程の検討を可能とするシステムとして、皮下移植により、肝臓、肺、脳、腎臓およぴ骨組織に高率で遠隔転移を来たす乳癌細胞株を樹立し、骨組織および軟組織双方への転移に対する検討できるシステムを確立した。このシステムを用いてビスフォスフォネートの効果をin vivoで検討した結果、ビスフォスフォネートは骨転移に対しては抑制的に作用する一方で、肺あるいは肝臓などの軟組織転移に対しては促進的に働くことが示唆された。
