高齢者の唾液腺における水輸送低下の分子機構とその治療薬の開発

研究概要

1.耳下腺・顎下腺に局在する水輸送体AQP5のcis-2-methylspiro-(1,3-oxathiolane-5,3')-quinuclidineによる管腔膜での増量とその加齢変化
8、52、104週齢ラットより耳下腺・顎下腺の切片を調製し、cis-2-methylspiro-(1,3-oxathiolane-5,3')-quinuclidine, pilocarpine, acethycholineやepinephrineとKrebs Ringer Tris (KRT)液(pH7.4)中で37℃にて15秒から30分間加温振盪した後、LongbottomらやVayroらの方法を応用して基底膜,管腔膜,細胞内小胞膜等に細胞分画する。SDS-PAGEに供した後、ニトロセルローズ膜に転写し、抗AQP5抗体を使ってウェスタンブロッティングを行い、デンシトメーターにて解析した。AChにより104週齢ラットの耳下腺管腔膜におけるAQP5は、AChにより15秒後に8週齢ラットと同様に最大に達した。しかし、増加率は8週齢ラットが3.5倍であるのに対し、104週齢ラットでは2倍であった。cis-2-methylspiro-(1,3-oxathiolane-5,3')-quinuclidineによっては、10分後に8週齢ラットが3倍であるのに対し、104週齢ラットでは2倍であった。cis-2-methylspiro-(1,3-oxathiolane-5,3')-quinuclidineはAChとは異なり、管腔膜でのAQP5増量が持続的であり、30分後に8週齢ラットで2倍、104週齢ラットでは1.8倍であった。
2.耳下腺・顎下腺の遊離細胞における分泌刺激薬による[Ca^<2+>]iの動態の加齢変化
各週齢ラットの耳下腺・顎下腺切片を0.5mMEGTAの存在下コラゲナーゼとヒアルロニデースで処理して、遊離細胞を得た。KRT液に浮遊後、Fura2-AM(10μM)を30分間室温にて負荷した。種々の作動薬と反応後、[Ca^<2+>]iを蛍光光度計にて測定した。cis-2-methylspiro-(1,3-oxathiolane-5,3')-quinuclidineによる[Ca^<2+>]iの上昇は104週齢ラットにおいても持続的であったが、ピーク時の上昇率は8週齢ラットに比し小さかった。
