口腔の知覚伝達系の発達障害に関する神経機能回復モデル構築

研究概要	培養神経細胞を用いて、細胞間のシナプス形成と相互作用、形成された神経回路網の情報伝達メカニズムを明らかにして、ヒト口腔の知覚伝達系機能障害の病態解明のための実験モデルを構築することを目的に研究を実施した。サーカディアン振動体としての機能が明らかにされている視交叉上核の細胞群を対象とし、マルチ電極皿(MED)の上で分散培養法、あるいは静置器官培養法による初代神経培養を行い、4〜8個の単一神経細胞の自発発火を2週間以上同時計測した。各々の培養条件で自発発火のサーカディアンリズムの同調が認められ、そのリズム同調はNaチャンネルブロッカーであるテトロドトキシンの長期投与によって阻害された。この結果は、細胞間のリズム同調の維持にとって、Naチャンネル依存性シナプス連絡が重要であることを示唆している。次に、腔からの知覚入力を受ける孤束核と、舌の運動神経が存在する舌下神経核を含む延髄神経核群での神経活動について、新生ラット(生後1-14日:P1-14)でのc-fos発現を指標として吸啜運動前後の発現量を比較した。吸啜運動の後では、孤束核と延髄網様体背側部にc-fos蛋白(Fos)陽性細胞の著しい増加が認められた。孤束核でのFos陽性細胞数はP5-7で最高となり、P14では減少したが、延髄網様体背側部でのFos発現に日齢による差は認められなかった。舌下神経核では吸啜運動後においてもFos陽性細胞はほとんど認められなかった。また三叉神経中間亜核では腹側部に少数のFos陽性細胞を認めたが、吸啜後の上昇はみられなかった。以上の結果から、吸啜期ラットの摂食行動には孤束核と延髄網様体背側部のニューロン活動が深く関与していることが明らかになった。今後、これら延髄ニューロン群の細胞培養系を確立し、視交叉上核ニューロン群と同様の解析方法により各々の神経核に固有な神経回路網の機能を明らかにしていく計画である。
