膵臓がん肝転移モデルの設立と治療薬開発への応用

研究概要	膵臓癌は治療の困難な癌であるが、その原因は原発巣が発見された時点で肝転移が生じていることが多いためである。したがって膵癌の治療を改善するには、 (1)肝転移が生じているかどうかを原発巣の遺伝子発現を解析することによって予想すること、(2)既に生じてしまった微小転移を薬物治療によって撲滅する方法を開発すること、の2点が重要となる。本研究では、膵癌の進行と肝転移形成能の上昇と伴う遺伝子変化を明らかにすることをめざし、DNAアレイ法によっていくつかの遺伝子を同定し、PCR法によって確認した。次にヌードマウスでヒト膵癌転移モデルを設立し、これを用いていくつかの薬物による微小転移治療の可能性を追求した。ASPC-1細胞、HPAC細胞、Capan-1細胞で脾注肝転移の系が樹立された。
