膵臓がん肝転移モデルの設立と治療薬開発への応用

研究概要

本研究の目的は、転移性の高いヒト膵臓癌に高発現または低発現している遺伝子を同定し、それらを膵臓癌の個性診断に利用することによって、膵臓癌の治療法を改善すること、これらの遺伝子の機能を調べることによって悪性度の高い膵臓癌の新しい治療法を開発することである。本年度に用いた細胞株は、COLO357FGからVezeridisらによって得られたバリアントであるが、設立後転移性に関する査定がなされていなかった。そこで、ヌードマウスを用いて同所移植(膵臓内移植)肝転移及び脾注肝転移のモデルで転移性を比較した。COLO357L3.5pl細胞及びCOLO357L3.5sl細胞共に、同所移植モデルで親株であるCOLO357FG細胞よりも高い転移性を示した。そこで、COLO357L3.5sl細胞とCOLO357FG細胞を用いて、遺伝子発現プロファイリング解析を行なった。発現が異なると判定された遺伝子は、さらにRT-PCRによってその発現レベルの差異を確認した。高転移バリアントでは,45遺伝子の内31遺伝子の発現が相対的に高いことが確認された。総体レベルが3倍以上のものに、神経細胞成長因子のレセプター及びそのシグナル伝達を行なう細胞内分子(p145TRK-B)が含まれていた。これらの分子の膵臓癌組織内における分布や患者間の相対強度の違いと,生存率の相関について、組織染色法によって解析中である。本研究によって、神経細胞成長因子のレセプター及びそのシグナル伝達分子の、膵臓癌の悪性挙動との関係が初めて示され、膵臓癌治療の分子標的としての有用性が示された。
