分子擬態を利用したRNA新薬の創製

研究概要

ランダムな配列を持つRNAプールから目的とする活性を有するRNA分子(アプタマー)を試験管内で濃縮・取得するSELEX法により、標的分子のアゴニスト、アンタゴニスト、リガンド等を作成しRNA性医薬品の開発へ応用する目的で、平成11年度はTc細胞の活性化において主要な役割を持つインターロイキン2(IL-2)分子および抗IL-2抗体を標的にしたSELEXを行った。試験管内レベルでIL-2および抗IL-2抗体に結合するRNA分子を複数作成することができたが、最終的に細胞レベルでの陽性クローンは得なれなかった。原因として、今回用いた条件が生体内と異なるため、細胞レベル/生体レベルでの評価に耐えなかった可能性が考えられる。また、中和抗体のepitopeのmappingを行う必要もあり、IL-2の受容体への結合様式をきちんと検討し、それに見合うような中和抗体の作製が必須となってくると思われる。これらの成績より(本質的にRNAとの親和性が薄いと思われる)サイトカインを標的とした戦略の見直しが必要と判断した。平成11年1月から新たに次の2つの戦略を研究に組み込み、推進した。1)翻訳開始因子(eIF)に結合する。RNA分子(アプタマー)の作成と、2)培養細胞にランダム配列の遺伝子を導入して生理的、形態的変化を引き起こすRNAアプタマーの検索を行う。eIFは真核生物において少なくとも30種類が知られており、以下の点でRNAアプタマーの作用を評価するモデルとして適している。すなわち、いくつかのeIFはRNA結合能があるためRNAアプタマーを得やすいこと、複数のeIFが高次構造をとることによって機能するためにアプタマーにより高次構造が崩されて機能が阻害されることが期待できることである。さらに幾つかのeIFは過剰発現により細胞の癌化を起こすため、作成したRNAアプタマーの癌抑制効果を試験し、医薬品化の可能性を検討できる。現在、1)についてはeIF-4G、eIF-4Aとうを標的にRNAアプタマーの作成を実施している。
