海洋生物付着阻害物質の探索および開発研究

研究概要	人工建造物、船底の付着生物による生物汚損を防ぐため、従来はスズ化合物などの重金属化合物を混ぜた塗料が多用されてきた。しかしスズ化合物は内分泌かく乱物質とされ使用がほぼ禁止されており、代替品の開発が望まれている。そこで本研究では環境への影響が小さいと期待される有機化合物に着目した。天然由来の生物付着阻害活性物質の探索を行うことで、環境にとって安全な生物付着防止剤の開発と、付着現象のメカニズムの解明を目的とした。まずアッセイ法として、付着性細菌Rhodospirillum salexigens SCRC 113株に対するペーパーディスクを用いた簡便な抗菌活性試験法を確立した。次いでカイメンや海藻などの抽出物に対しスクリーニングを行い、活性が認められた種の分離を行った。沖縄県にて採集したカイメンPseudoceratina sp.からは新規ブロモチロシン類縁体、ザマミスタチンを単離し、その構造を決定した。ザマミスタチンは特に強い抗菌活性(1.6μg/disc,阻止円21mm)を示した。またカイメン Terpios sp.からはナキトリオールを単離した。ナキトリオールは比較的簡単な構造の化合物であったため合成を行い、量を確保した。現在、野外試験を含む各種活性試験を検討している。付着阻害活性を示すトリブロモアセトアミド類に関しては、各種誘導体の合成を行い、構造活性相関を検討した。N-ベンジル-N-メチル体など数種の誘導体には、付着阻害活性に加えて腫瘍細胞に対する細胞毒性やチューブリン重合阻害活性も確認された。現在付着阻害に関する野外試験を行っており、実用性を検討している。
