研究概要

1.ハンタウイルス血清型鑑別抗体測定用抗原として、バキュロウイルス発現ベクターによってハンターン型、ソウル型、ドブラバ型の核蛋白抗原の、血清型交差エピトープ(N末端50アミノ酸)を除いた抗原を作成し、ELISA抗原として応用した。それぞれの血清型ウイルスに感染した患者血清を用いた結果、中和試験と同等に血清型鑑別が可能であることが明らかとなった。また、極東ロシアと中国で得られた患者とげっ歯類血清を用いてさらに有用性を検討中である。しかし、北欧流行ウイルスであるプーマラ型ウイルスの核蛋白コード遺伝子のクローニングと発現が未だ成功していないため今後の課題である。
2.リンパ球性脈絡髄膜炎(LCM)ウイルス国内分離株(OQ28)及びWE株(プロトタイプ)遺伝子の核蛋白コード領域から得られたPCR産物をTAクローニング後、発現ベクターへの置換し、全長、C末領域と中間領域の3つの領域について哺乳類細胞用発現ベクターを作出し、赤色蛍光を発するCOS細胞を得た。この核蛋白コード領域に由来する抗原の大量発現をめざし、pIVEXベクターに導入し、RTS500システムを用いた発現を現在行っている。
3.LCMウイルス核蛋白を発現させたバキュロウイルス感染細胞(Sf-9細胞)を用いて、組み換えELISAを確立し、またIFA抗原としてTm5細胞抗原を作製した。両抗原の評価を、わが国のマウス動物実験施設、延べ1,117施設由来9,840検体を対象に実施した。しかし、ELISAにおいては65施設(5.8%)、119検体(1.2%)において陽性反応が認められたしかしIFAによる確認試験において、それらすべて陰性であり、ELISAにおける陽性反応は偽陽性反応であると考えられた。このことよりELISAは大量検体処理のためのスクリーニング試験に、IFAはその結果の確認試験に有用であることが示された。
