高機能性キメラコラーゲンの作製

研究概要	上皮増殖因子(EGF)とコラーゲンとを融合したタンパク質を作製し、この分子の有用性を検討した。得られた組換えタンパク質をイムノブロット法により解析した結果、抗EGF抗体と抗コラーゲン抗体の両抗体と反応するタンパク質を検出した。また、EGF-PROのコラーゲン部分がコラーゲンに特徴的な三重らせん構造を有していることをペプシン消化実験により確認した。更に、EGF-PROの細胞増殖活性を、EGF依存性のBalb/MK-2細胞を用いて調べところ、EGF-PROはキメラ化していないEGFとほぼ同等の細胞増殖活性を持っていることが明らかになった。キメラ化によってコラーゲンの立体構造およびEGFの細胞増殖活性が影響を受けなかったものと考えられる。またEGF-PROは通常のコラーゲンと混ぜることでディッシュ上に固定可能であった。本研究によりEGFとコラーゲンをキメラ化することによりEGF活性をディッシュ上に固相化出来るようになった。コラーゲンゲルを用いた徐放システム開発を目指すためインターロイキン2(IL-2)とコラーゲンとの融合タンパク質を産生した。イムノブロットにより、IL-2とIII型コラーゲン(3A1)のキメラタンパク質が産生されていることを確認した。また、IL2-3A1のコラーゲン部分がペプシン耐性の三重らせん構造を形成していることを確認した。更に、IL2-3A1のIL-2部分がキメラ化していないIL-2とほぼ同等の細胞増殖活性を有していることをマウスT細胞由来のCTLL-2細胞を用いて確認した。in vitroモデル系の実験を行い、コラゲナーゼによって遊離したIL-2部分が細胞増殖活性を保持していることを確認した。以上のことから、IL2-3A1はコラーゲンの生分解に伴いサイトカインを徐放するドラッグデリバリーシステムを構築するのに有用であると考えられる。
