研究概要	DNA(mRNA・タンパク質)の固定化と選択回収技術:核酸プローブを表面に固定した基板を用いて、溶液中の試料DNAを高速かつ高分解能に分離・精製することを目的とする遺伝子前処理技術を検討した。ガラス基板表面にクロムを6nmの厚さに蒸着し、それをシラン化処理することによって核酸プローブを固定して核酸プローブ基板とした。試料核酸プローブの方に蛍光色素を結合して蛍光画像観察したところ、単なるガラス基板と比べて核酸プローブが均質にむらなく付着することが分かった。そこで、このクロム蒸着面に近赤外レーザー集束光を顕微鏡下で照射することによって、クロム表面を局所加熱し、μmオーダーの空間分解能でDNAを熱変性し、試料核酸プローブを局所分離することに成功した。この試料核酸プローブの熱解離は可逆的で、再結合に関しても変性することなく非常に良い再現性を示した。また基板の蒸着クロムを面電極としてDNAを基板表面に誘導することができた。また、異なるプローブが付着した複数の領域を基板上に2次元に配置し、試料DNAが特定の領域にしか結合しないこと、また、他の領域に影響を与えることなく特定の領域に結合したDNAのみを回収できることを確認した。Caged DNAの合成:DNA塩基のアミノ基を化学修飾(Cage化)することによって複製機能を失活し、紫外光照射によってCageを外して再活性化させることのできる不活性化DNA(Caged DNA)を合成することを試みた。その結果、光照射による再活性化がある程度可能であることがPCR法を用いて確認された。以上2つの技術は、本研究課題を推進する上での鍵技術になることが期待される。
