昆虫で初めて同定されたサイトカイン・その活性化機構と多機能性に関する研究

研究概要

発育阻害ペプチド(Growth-blockingpeptide, GBP)は、昆虫の発育を調節する生理活性ペプチドとして同定された。最近の研究により、GBPは昆虫の発育調節のみならず、細胞増殖活性や血球活性化作用などの多機能性を有するサイトカインであることが明確になった。さらに、GBPは、主に脂肪体や中枢神経組織で分子量約1.5KDaの前駆体の形で合成されたプロセッシングを受けて活性化することが明らかになっている。
本研究においては、主にこの活性化に関与するプロセッシング酵素に着目し、さらに、限定加水分解によって活性化された後のGBPの細胞性免疫作用への関与を考察することが当初の目的であった。ProGBPプロセッシング酵素の単離については、GBP発現量の多い脂肪体を精製出発材料に用い疎水カラムクロマトグラフィー,ゲルろ過カラムクロマトグラフィー,イオン交換カラムクロマトグラフィー、最後にもう1度ゲルろ過カラムクロマトグラフィーという全部で4段階のカラムクロマトグラフィーによって精製を完了した。精製されたプロセッシング酵素の分子量は、約38KDaと計算された。精製されたこの酵素が間違いなくGBP前駆体プロセッシングに関わっていることは、前駆体GBPと反応させることによってGBPのアミノ末端アミノ酸の前で限定加水分解が起ることから証明した。
また、各種合成ペプチドを基質に用いてその特異性を調べたところ、哺乳動物の血液凝固反応に関与するファクターXaに似たセリン型プロテアーゼであることが明らかになった。一般的に昆虫においては血液凝固反応がほとんど見られない。今回精製されたproGBPプロセッシング酵素が一次構造の面でもファクターXaに類似性をもち、さらに、昆虫サイトカインのプロセッシングに関与していることが明らかになれば哺乳動物の血液凝固系の分子進化を考える上で特に興味深い。
