研究概要

本研究は、細胞表層糖鎖モデルとして一連の人工糖鎖高分子を作製し、糖鎖-糖鎖間や糖鎖-タンパク質間の相互作用の起源を分子レベルで解明することを目的としている。
1.リガンド糖鎖の酵素合成
糖鎖-糖鎖間の認識におけるリガンドオリゴ糖鎖のコア4糖として知られているラクト-N-テトラオースおよびてラクト-N-ネオテトラオースを出発基質としてこれらを糖転移酵素によるフコシル北を行って糖鎖レセプターの生物活性エピトープ(抗原決定基)として知られているLeXオリゴ5糖およびこれらのアナログであるオリゴ糖鎖の実践的酵素合成法を確立した。
2.細胞表層糖鎖モデルの構築
細胞表層の糖鎖の機能的シミュレーションを目指し、疎水的高分子の骨格を分子基盤として、これに糖残基を導入した細胞表層のクラスターモデルを系統的に構築を行った。即ち、ポリグルタミン酸の側鎖にN-グリコシル化反応を応用して上記オリゴ4-5糖鎖を導入し、糖鎖一ポリペプチッド複合体を作製した。この糖鎖高分子を細胞表層のモデルとして設定し、糖鎖の機能的シミュレーションの解析が進行中である。
