変異酵素と合成基質プローブを用いた大腸菌ユビキノール酸化酵素の構造と機能の解明

研究概要	大腸菌末端酸化酵素(bo型ユビキノール酸化酵素)のユビキノール酸化部位の構造及び機能特性を解明する目的で、系統的に構造修飾した一連のユビキノール類縁体の構造活性相関研究を行った。側鎖構造の如何に関わらず、2位エトキシ体の活性が3位エトキシ体よりも顕著に低下したことから、キノール環上の2位メトキシ基は3位メトキシ基よりも酵素によってより厳密に認識されていることがわかった。また、5位メチル基は酸化反応に重要な役割を果たしていることがわかった。bd型酵素についても同様の解析を行い、基質認識がbo型酵素と類似している事がわかった。次に、ユビキノール酸化部位に作用する置換フェノール系阻害剤に薬剤耐性を獲得した2種類のbo型変異酵素を用いて、一連のユビキノール類の電子伝達活性を評価し、野生型酵素における電子伝達活性と比較検討した。ここで用いた薬剤耐性変異酵素の変異部位は、サブユニットIIの親水性に富んだC末端(Ser258およびGln233)に位置していることがわかっている。ユビキノール類の構造活性相関プロフィルを野生型酵素と薬剤耐性酵素で比較したところ、意外にも全く差違が認められなかった。このことは、置換フェノール系阻害剤に耐性を与えるような上記の構造変化は、基質認識そのものには影響を与えないことを示唆している。続いてbo型ユビキノール酸化酵素のユビキノン結合ドメインを光親和性標識法によって同定するために、光分解性ユビキノン(2-アジド-Q2および3-アジド-Q2)の合成方法を確立した。
