研究概要

強力な発がんプロモーター・ホルボールエステルは、細胞内情報伝達の鍵酵素であるプロテインキナーゼC(PKC)を主要なターゲットとしている。ホルボールエステルは、PKCの調節領域に2個連続して存在するアミノ酸残基約50個からなる亜鉛フィンガー様配列(C1AおよびC1Bドメイン)に結合する。脳および神経系にのみ存在するPKC gammaは、C1AおよびC1Bドメイン共に強力なホルボールエステル結合能を有する特異なアイソザイムである。PKC gammaのC1ドメインの機能を明らかにし、本酵素の活性化を選択的に制御できる化合物をデザインする目的で、PKC gammaの2つのC1ドメインを含む116残基からなるモデルペプチド(gamma-C1A-C1B)を化学合成した。本ペプチドは、Fmocアミノ酸およびHATUを用いた固相法によりフラグメント縮合を用いることなく一気に合成でき、複数の逆相系HPLCにより高純度で得られた。MALDI-TOF-MSおよびトリプシンによるペプチドマッピングによりその構造を確認した。化学合成によって得られたペプチドとしては、最長の部類である。亜鉛でフォールディングしたgamma-C1A-C1Bは、全長のPKC gammaと同等のホルボールエステル結合能を示し、エレクトロスプレー質量分析法により、亜鉛を4原子配位していることを明確に示すことができた。本モデルペプチドを用いて、各種ホルボールエステルのダイマーの結合能を調べた結果、炭素数10-12個のスペーサーを持ったダイマーが特に強く結合することが明らかになった。
