絶食ストレスによる性腺機能抑制におけるエストロジェンフィードバックの脳内機構

研究概要

これまで48時間の絶食ストレス負荷により、視床下部室傍核(PVN)および延髄孤束核(NTS)においてエストロジェン受容体α(ERα)の発現が誘起されることを明らかとし、またこれらの神経核が、絶食ストレス不可による黄体形成ホルモン(LH)分泌抑制に必要なエストロジェンのフィードバック部位であることを明らかにした。そこで、PVNでのERα発現がどのような機序により誘起されるかを解明し、さらにNTSにおけるERα発現ニューロンを同定することを目的として、ラットを用いて実験を行った。
卵巣除去を施したラットを用い、48時間の絶食を負荷した。絶食開始より、6、24、30および48時間後に脳を採取し、免疫組織化学によりERαの免疫陽性細胞の有無を確かめた。これに加え、一部の脳切片において、ERαとチロシン水酸化酵素(TH)あるいはドーパミンβ水酸化酵素(DBH)との2重免疫組織化学を行った。また、グルコース拮抗剤の投与によるERα発現の変化の有無も同様の方法を用いて確認した。
この結果、PVNおよびNTSにおけるERα免疫陽性細胞の増加は、絶食開始30時間以降に認められた。また、PVNにおいてERα免疫陽性細胞の殆どが、細胞質にTHあるいはDBHを持つことが明らかとなった。さらに、PVNにおいて、ERα免疫陽性細胞に隣接して、THあるいはDBH免疫陽性繊維が認められた。またグルコース拮抗剤の投与によっても、同様のERαの上昇が確認された。
以上の結果から、絶食はNTSのA2領域に細胞体をもつノルアドレナリン作働神経におけるERαの発現を促すこと、およびPVNにおけるERα免疫陽性細胞は、ここに投射するノルアドレナリン作働性神経により、ERα発現が誘起されることが示唆された。絶食によるさまざまな生理変化のうち、グルコース利用性の低下が、この発現誘起に関わることが示唆された。
