時計遺伝子の機能:視交叉上核培養系における検討

研究概要

本研究では、視交叉上核神経細胞のサーカディアンリズムを指標に、個々の時計遺伝子や候補遺伝子の機能を明らかにすることを目的として、自家繁殖したWistar系ラットおよびリズム変異マウスClockを用い、以下の実験を行った。
(1)アンセンスによる時計遺伝子機能の検討
マルチチャンネル細胞外記録システムを用いて新生仔視交叉上核の分散培養を行い、単一神経細胞の自発発火リズムを連側測定し、安定したサーカディアン周期のリズムを発振する時計細胞において、その様々な位相に時計遺伝子BMAL1のアンチセンス10μMを6時間投与し、その後の周期変化、位相変位を解析した。その結果、個々の視交叉上核細胞においてリズム位相依存性の位相変位を確認した。アンチセンス効果は、免疫組織学的検索可能な高感度の抗体が作成できず、in situ hybridizationによるmRNA発現レベルでの判定に止まった。この結果、BMAL1遺伝子がリズム発振に必須の遺伝子であることが単一視交叉上核神経レベルで明らかとなった。
(2) Clock変異マウスの視交叉上核神経細胞活動リズム
Clock変異マウスの視交叉上核をマルチ電極上でスライス培養し、神経活動リズムを長期にわたり測定解析した。その結果、単一視交叉上核神経細胞の活動リズムの平均周期は、ホモ接合マウスで27.2時間、へテロ接合で24.8時間、野生型では23.5時間であり、行動リズムの周期とほぼ一致した。また、神経活動リズムの振幅、リズムを示す神経の割合には野生型とClockマウスとの間に有意差がみられなかった。Clockマウスの行動リズムは連続暗で無周期を呈するのに対し、単一神経レベルでは安定したリズムが出現することが明らかとなり、時計遺伝子Clockの変異は視交叉上核レベルではサーカディアン周期を延長させ、行動リズムの無周期はリズム出力の障害であることが分かった。
