研究概要	視交叉上核におけるPer1遺伝子の誘導の限局性と時差ボケの機序の解明夜間の光照射によってラットPer1は視交叉上核の腹外側部に発現し背内側部には誘導されなかった。Per1の誘導は位相変位のinitial stepと考えられており、このようにPer1遺伝子が限局した部位にのみ発現することは光反応性の時計と光には直接反応しない時計の2種類の体内時計が視交叉上核の中で乖離して存在することを示唆する。更に明瞭に2つの時計の存在を明らかにするため、急激な明暗サイクルのシフト後、視交叉上核の時計の位相をPer1の発現を指標として観察した。この際に背内側部の位相は明暗サイクルのシフト後にもとの位相からシフト後の位相に変位するのに約一週間を要しこれが時差ボケ現象の原因であると考えられた。遺伝子導入ショウジョウバエを用いた哺乳類PER1タンパクの機能解析ショウジョウバエの時計遺伝子ピリオド(Per)のマウス相同遺伝子mPer1,mPer2をショウジョウバエのper、timelessプロモーターの制御下においてショウジョウバエに導入し、行動の変化について検討した。mPer1,mPer2を導入したピリオド遺伝子欠失体(Per0)のうち、いくつかの系統にて、概日リズムの回復を認めた。マウス時計遺伝子導入トランスジェニックマウスの解析 Per1,Per2,Per3 cDNAおよび、おのおのの遺伝子の上流の調節領域を採取し、そのプロモーター活性を測定した。これらのcDNAを発現ベクターに組み込み、エピ卜-プタッグとしてFLAGを下流に結合し、株化細胞であるCOS7に発現させたところ,Western blottingにてタンパクの発現を検出できた。また、GFPにPer1,Per2,Per3を結合させて、その発現を観察した。これを用いてトランスジェニックマウスの作成段階に入っている。最近になり他施設よりPer1を過剰発現するトランスジェニックラットが作成された。今後、このようなラットに関しても時計遺伝子の発現の解析を行う予定である。また、TetracyclinによってPer1の発現が誘導される遺伝子導入動物の作成にも着手している。
