活性酸素種のよる細胞表面glycocalyxの修飾と細胞接着性の変化の検討

研究概要	細胞接着能は細胞表面の接着分子の変化によって左右され、炎症や癌転移の際の細胞の接着に影響を及ぼすと考えられ、一方活性酸素種(ROS;Reactive Oxygen Species)は炎症や癌の局所で多量に生成することが明らかになっている。今年度は活性酸素と細胞接着について以下のことを明らかにした。まず、ヒト臍帯より血管内皮細胞を単離し、第4〜8継代の細胞を用いた。シクロヘキシミドならびにアクチノマイシンD処理下において血管内皮細胞に対して5分間のROS処理(過酸化水素、ヒポキサンチン+キサンチンオキシダーゼ、一酸化窒素など)を行い、癌細胞の接着性を検討した。癌細胞は大腸癌由来のColo201をBCECF-AM(3'-O-Acetyl-2',7'-bis(carboxyethyl)-4 or 5-carboxyfluorescein,diacetoxymethyl ester)で蛍光ラベルし、ROS処理後10〜80分まで接着性を観察した。その結果活性酸素処理内皮細胞への癌細胞の接着は処理後経時的に増加し、その程度はヒポキサンチン+キサンチンオキシダーゼ群で大きかった。またこの接着性の亢進はSOD+カタラーゼもしくはEDTAによって抑制され、活性酸素の中でもヒドロキシルラジカルが接着性の亢進に作用していると考えられた。またヒト末梢血より単離した好中球をBCECF-Amで標識して用いた実験でも同様の結果が得られた。以上より新規蛋白の合成阻害下において細胞接着は亢進しており、このことより接着分子の発現誘導ではなく活性酸素によって接着が亢進することが考えられた。すなわち炎症や癌転移に際して好中球由来の活性酸素や化学療法、放射線療法由来の活性酸素によって血管内皮細胞表面が直接修飾され、細胞接着が亢進する新たなメカニズムの存在が示唆された。
