新規痛覚制御ペプチド・ノシスタチンの受容体及び産生機構の解明

研究概要	我々は、オピオイド受容体μ、κ、δの類縁体受容体の内因性リガンド、ノシスタチン/オーファニンFQ(Noc/OFQ)の前駆体蛋白上に、Noc/OFQの痛覚反応を抑制する生理活性ペプチドを見いだし、ノシスタチンと名づけた。ノシスタチンは、Noc/OFQ受容体には結合せず、別の受容体の存在が示唆された。本研究は、ノシスタチンとNoc/OFQのペプチドの産生制御機構やノシスタチン受容体の解明を目的とした。まず、産生機構の解明のため、ラジオイムノアッセイ系を確立し、ペプチドの単離、同定、定量を行った。ウシノシスタチンは、17個のアミノ酸のぺプチドであるが、ヒト、マウス、ラットでは、30、41、35アミノ酸のペプチドとして存在し、ヒト慢性痛患者の髄液においてもノシスタチンの存在が認められた。いずれのノシスタチンもNoc/OFQによる痛覚反応を抑制し、この効果は種間で保存されているC末端6つのペプチドのみで引き起こされた。Noc/OFQとノシスタチンのペプチドはともに視床下部に最も多く存在し、免疫組織染色法により、ノシスタチンは脊髄後角の表層、脳においては三叉神経脊髄路、傍正中縫線核や視床下部の腹内側核に存在しており、Noc/OFQ局在と一致していた。さらに、ノシスタチンは、プロスタグランジンE_2により誘導される痛覚反応やホルマリンによる末梢性炎症の痛みに対しても抑制効果を示した。海馬において、Noc/OFQのグルタミン酸の遊離の抑制効果が、ノシスタチンによって抑制された。ノシスタチンが記憶、学習障害を改善する報告がなされ、Noc/OFQの中枢作用に対し、ノシスタチンがどのように関与しているのか興味ある点である。今年度の研究によりノシスタチンの存在とその役割が明らかとなったが、平行して、現在ノシスタチンとNoc/OFQプロセッシングに関与する酵素の同定や受容体の解明を進めている。
