ヒト肺癌におけるp27^<KIP1>の発現調節異常の分子機序とその治療への応用

研究概要	昨年度までに小細胞肺癌におけるp27^<KIP1>高発現の分子機序の解析を行ない、ほぼ全例においてp27^<KIP1>が高い発現をしていること、低栄養及び低酸素状態などの細胞環境の悪化に伴ってp27^<KIP1>が誘導され、G1/G0期に細胞周期を留め、アポトーシスを防ぐ役割を果たしていることを示唆する結果を得た。本年度は、将来の臨床応用を視野において、p27^<KIP1>が強く誘導される典型的な細胞株に対するp27^<KIP1>の合成アンチセシスDNAオリゴマーのアポトーシス誘導効果について検討した。 p27^<KIP1>が強く誘導される典型的な細胞株ACC-LC-48に、3nMから10nMのC、U-プロピン化/チオール化アンチセンスDNAオリゴマー(15-mer)をカチオン化リポゾーム法によって導入したところ、約70%の細胞へオリゴマーの導入が見られた。この細胞をイソロイシンを含まない培地で培養するとオリゴマー濃度依存的にp27^<KIP1>の蓄積の低下が見られ、アポトーシスに陥る細胞の割合が増加した。一方、同様の塩基組成を持つミスマッチオリゴマーにはこの様な効果は認められなかった。また、イソロイシンを含む通常の培地で培養した場合にはアンチセンス及びミスマッチオリゴマーのいずれの場合にもアポトーシスの増加は見られなかった。以上の結果から、p27^<KIP1>に対するアンチセンスDNAオリゴマーによる肺小細胞癌治療への応用の可能性が示唆された。in vivoの腫瘍に対するにオリゴマーの導入効率の向上、及び他の治療法との併用による効果増強に至適な導入時期の選定などが今後の課題である。
