ヒト肺癌における細胞周期関連因子の異常の解析、並びに新たな診断、治療法への応用

研究概要

13年度は、1)ヒト肺癌の手術材料での臨床病理学的検索、2)昨年度までに当研究室が樹立したヒト肺癌培養細胞株、既に樹立され広く使用されている血球系、骨肉腫細胞株等を用いた系でのcyclin, cdkの潜在的機能解析を行った。
1-a)ヒト肺癌150例の免疫染色の結果、cyclin A陽性例は、どの組織型でも予後不良であったが、cyclin E陽性率は腺癌においては組織学的分化度と逆相関し陽性例は予後不良、扁平上皮癌では分化度と相関し予後良好であった。これは、cyclin Eの発現量は、腺癌ではそれが活性型の核内蛋白として免疫染色に反映されるが、扁平上皮癌では低悪性度の、発現レベルの低い癌でも分解能の低下のため多量に蓄積し、免疫染色で陽性となるためであった。大細胞癌は両方の染色パターンが見られ、非常に低分化の扁平上皮癌、腺癌の集合であることが示唆された(2002年、第91回病理学会発表予定)。
1-b)肺癌のみならず軟部肉腫でも、cdk-innibitor(p21,p27)は一定量の存在でcydin-cdk複合体を活性化し、その活性は腫瘍の悪性度を規定する事を明らかにした。(Am. J. Pathol. 2000,Int. J. Cancer 2001)。
2-a)肺小細胞癌7株、我々が樹立した小細胞癌、大細胞神経内分泌癌、血球系、骨肉腫の細胞株(Hela, Saos2)を用いて種々のcydins, cdkの潜在機能を検索した結果、どの株でもcdk4, cyclin D1の過剰発現により、細胞はアポトーシスに陥り、細胞の普遍的なアポトーシスのメディエーターである事が予想された。(2001年、第60回日本癌学会、第24回日本分子生物学会発表)
2-b)RB蛋白質が不活化した細胞では、cdk4/cydin D1のみならず、どのcydin, cdkの過剰発現によってもアポトーシスが誘導され、RB蛋白質はcdk4/cydin D1以外の蛋白によるアポトーシス誘導を抑制していることが示唆された(第60回日本癌学会、第24回日本分子生物学会発表)。
1)の結果は、診断上の付加価値を加える臨床病理学的意義と組織型による蛋白分解の機序の差異を明らかにした細胞生物学的意義を有し、2)の結果はcdk/cyclinによるアポトーシス誘導、またそこでの癌抑制遺伝子の関与等、将来の遺伝子治療のための貴車なデータとなり得ると考えている。
